In this study, three different fromulae of media containing some of the requirements needed for the production of tetanus toxin were used. The first formula was without any amino acids or vitamins which showed the least amount of tetanus toxin produces (5-10Lf/ml). The second formula of medium contained two amino acids which gave a good yield of tetanus toxin (30-40 Lf/ml).

While the third formula of medium with two amino acids and six vitamins showed a high yield of tetanus toxin (50-60Lf/ml).
The first fromula was excluded as the toxin producer did not comply the minimum requirements needed for use in production of tetanus toxin and hyperimmune serum. The toxin produces by the second or third formulae was suitable for use but formula III was more complicated and highly expensive.

So it is advisable to use the second fromulae which comprised only two amino acids (cystine and tyrosine ) as a simple and more economic medium for production of tetanus toxin, The produced toxin showed a good immunogenicity which gave a high titer of antitetanic serum (400 IU/ml) after five injections in horses within a short preiod of intervals.
