RT-PCR of Gi Gene of Rift Valley Fever Virus

Summary
 


Eighty percent cytopathic effect (CEP) was developed on monolayer Baby Hamster Kidney (BHK) cells 36 hours post inoculation with Rift Valley fever virus (RVFV) ZH-501 Using QIAGEN Rneasy Mini Kit, the infected cells were lysed and the viral genome was extracted and purified.

 
 
 


Two primers were designed for the GI glycoprotein gene on the M. secgment of the RVFV (The Egyptian isolate). QIAGEN one step RT-PCR kit was utilized to generate full length copies of cDNA corresponding to the aforementiond gene and subsequently be used as a template in the proper polymerase chain reaction.

 
 
 


The length of the gene product was about 1440 bp. Getting the GI gene in a pure form will gave the way to further speculations aiming to the disease controlling.
