A polymerase chain rection (PCR) was developed and applied for the detection of the haemolysin gene (alpha toxin) of Clostridium Sepetion. The PCR primer were designed from the sequence of the haemolysin gene and synthesized.

The PCR results indicated that a fragment of 270 bp was obtained from refercence, vaccinal and 5 isolated strains of Clostridium. septicum, but not from strainas of Clostridium charavoei. When the PCR products was digested with Sau3 Ai, two DNA fragments of the expected 148 bp and 122 bp were recognized.

The PCR assay was sensitive (limit of detection was 2x10 cells per PCR), specific (no amplification products were obtained with other clostridia) and rapid as a good alternative to conventional methodologies for routinely screening a large number of samples. |
